- Is a talented writer and long time partner of Danny Tripp.
- Danny Tripp — played by Bradley Whitford
- Recently lost a job due to testing positive in a drug test. Together he and Matt Albie are asked to take over “show running” duties for a live, weekly, sketch comedy show.
- Jordan McDeere — played by Amanda Peet
- Has just been promoted to president of the show’s network: The National Broadcasting System. She takes charge after the producer of the show gives a speech live on the show about how they had been censored by the network. To solve the crisis she decides to hire Danny and Matt.
- Harriet Hayes — played by Sarah Paulson
- One of the main stars of the fictional “Studio 60″ show. She and Matt have had a past romantic relationship.
- Tom Jeter played by Nate Corddry
- Another of the show’s main stars.
- Simon Stiles played by D. L. Hughley
- The third main star. He attended the Yale School of Drama. Tom and Simon are united by their “mutual respect, their dedication to their show and nothing else”.
- Jack Rudolph played by Steven Weber
- Chairman of the network NBS and Jordan’s boss. He fired Matt and Danny from “Studio 60″ four years prior to the pilot and reluctantly allows Jordan to rehire them.
- Cal Shanley played by Timothy Busfield
- The control-room director of “Studio 60″. (Busfield will also serve as producing director on the show.)
